We are a community theater group looking for our next musical, need suggestions we usually have 40-50 in the show which includes chorus. We have already done, Willy Wonka, Wizard of OZ, Annie, Music Man...

Seussical -- a little more current, but very familiy friendly, very fun music, the lead is spread out among a half-dozen principals, and there's room for as many Whos and/or Jungle Animals as you want.

Look at Anne of Green Gables - The Musical from Samuel French (there are a number of other versions out there, but this is the original). This is a standard book musical, and there are good roles spread out through all age groups. It is a well know name that should draw well.

YouthPLAYS publishes The Secret Garden by Miriam Raiken-Kolb, a new musical version of the classic story with potential for a large cast, and obviously the title has name recognition. Definitely worth checking out:
